Supplemental Material: Analysis of Endocrine Disruption in Southern California Coastal Fish Using an Aquatic Multispecies Microarray

چکیده
Estradiol and 4-nonylphenol exposure studies in zebrafish. Approximately eighty male zebrafish were divided evenly between three 80 L glass aquaria. Each aquarium was individually heated to maintain a temperature of 26 /29 °C with a light-dark cycle ratio of 14:10 h. Fish were acclimated for one week prior to commencing the experiment. One tank contained only tap water (negative control) and the others contained water with either 10 M estradiol or 4-nonylphenol. All exposures utilized a continuous flow-through system, to maintain constant concentrations of the chemicals during the two-week experiment, after which the fish were anaesthetized with 3-aminobenzoic acid ethyl ester (10 g/L) and liver samples harvested, frozen in liquid nitrogen and stored at –70 °C.

Analysis of Endocrine Disruption in Southern California Coastal Fish Using an Aquatic Multispecies Microarray
BACKGROUND Endocrine disruptors include plasticizers, pesticides, detergents, and pharmaceuticals. Turbot and other flatfish are used to characterize the presence of chemicals in the marine environment. Unfortunately, there are relatively few genes of turbot and other flatfish in GenBank, which limits the use of molecular tools such as microarrays and quantitative reverse-transcriptase polymera...
متن کاملMolecular Analysis of Endocrine Disruption in Hornyhead Turbot at Wastewater Outfalls in Southern California Using a Second Generation Multi-Species Microarray
Sentinel fish hornyhead turbot (Pleuronichthysverticalis) captured near wastewater outfalls are used for monitoring exposure to industrial and agricultural chemicals of ~ 20 million people living in coastal Southern California. Although analyses of hormones in blood and organ morphology and histology are useful for assessing contaminant exposure, there is a need for quantitative and sensitive m...
متن کاملIncreased sensitivity in the detection and quantitation of virulence factors of Aeromonas hydrophila and Aeromonas caviae using quantitative PCR
Aeromonas spp., opportunistic pathogens of humans and fish, are ubiquitous in the aquatic environment. Aeromonas hydrophila and Aeromonas caviae have increasingly been isolated from patients with gastroenteritis and superficial wound infections and the former also has been associated with hemorrhagic septicemia in fish. This is the first study to report Aeromonas occurrence and frequency of vir...
متن کاملEvaluation of reproductive endocrine status in hornyhead turbot sampled from Southern California's urbanized coastal environments.
As part of a regionwide collaboration to determine the occurrence of contaminants and biological effects in coastal ecosystems offshore of urban southern California, the present study characterized the reproductive endocrinology of an indigenous flatfish, the hornyhead turbot (Pleuronichthys verticalis), and compared groups sampled from different study sites representing varying degrees of poll...
متن کاملIntegrated coastal effects study: synthesis of findings.
Municipal wastewater discharges constitute a major source of contaminants of emerging concern (CECs) to coastal waters, yet uncertainty exists regarding their linkage to adverse biological effects such as endocrine disruption. Limited information is available concerning the types and fate of CECs discharged to the Southern California Bight (SCB) from municipal wastewater and their potential for...
